I couldn't answer with just one movie especially considering different genres. There are just too many for me to choose. The Godfather is definitely up there.

Star Wars, this film has inspired many things in my life from my hobbies to reading sci fiction books to taking up a science degree at university to my current hobby of kendo and iaido.
And no matter how many times i watch it, I still get the goose bumps when that giant star destroyer comes over head.

Wrath Of Kahn is right up there. Spock dieing is the downside.
Alien for its shock value. What makes it scary is it's almost believable.
