I'm adding 32mg of memory to my HP printer and can get 2 SIMM modules of 16mg or 1 single 32mg module.

They are the exact same price and I have 2 slots available so I can live with either configuration.

Is there any advantage for 1 over the other?

yes
the 32 leaves an empty slot for the next upgrade.

There will be no further upgrades. I'll have 48mg in my HP 5P and that's all I'll ever need. It only came with 2mg when I bought this gem almost 9 years ago. It only has 4270 pages through it since new.

Besides the space availability is there any other advantage to 1 32mg over 2 16mg?

The only advantage I can see is if you have 2 and one happens to go bad you can still use it with one until you get a replacement. Other than that it's a wash.
